鼻敏感方对变应性鼻炎模型大鼠血清细胞因子的调节作用

中文摘要:   目的:观察实验性变应性鼻炎大鼠血清白细胞介素-4(IL-4)、干扰素-γ(IFN-γ)、白细胞介素-17(IL-17)、白细胞介素-10(IL-10)的变化,探讨鼻敏感方对这4种细胞因子的调节作用及其治疗变应性鼻炎的可能机制  方法:32只SD大鼠随机分为空白对照组、模型组、鼻敏感方组、氯雷他定组。除空白对照组外,其余各组以卵清蛋白为致敏原建立大鼠变应性鼻炎模型,造模成功后分别给予0.9%NaCl溶液、鼻敏感方水提剂、氯雷他定水溶液灌胃。观察给药前、给药7天后、14天后、28天后大鼠行为表现,28天后采用酶联免疫吸附法(ElISA)检测各组血清中IL-4、IFN-γ、IL-17、IL-10细胞因子水平  结果:给药7天后、14天后、28天后,鼻敏感方组及氯雷他定组较模型组大鼠鼻部症状明显减轻(P<0.01)。与空白对照组比较,模型组IL-4、IL-17水平升高,IFN-γ、IL-10水平降低(P<0.01);与模型组比较,鼻敏感方组及氯雷他定组IFN-γ、IL-10水平升高,IL-4、IL-17水平降低(P<0.01)  结论:鼻敏感方是治疗变应性鼻炎的有效方剂,其治疗机制之一是通过影响IL-4、IFN-γ、IL-17、IL-10水平从而调节Th1/Th2和Th17/Treg细胞间平衡实现的。
Abstract:  Objective:The current research is to observe the contents change of IL-4,IFN-γ,IL-17 and IL-10 in allergic rhinitis rats’ serum,analyze the regulating role of Bimingan Prescription in the cytokines and the possible mechanism of its treatment of allergic rhinitis  Methods:32 SD rats are randomly divided into blank group, model group, Bimingan group and Loratadine group. Apart from the blank group,other groups established AR rats model with OVA. When the model was established, fed the objects with normal saline, water extrat of Bimingan Prescription and Loratadine water solution. Observe the scores of rats symptoms for 7 days, 14 days and 28 days after afterwards. Detect cytokine of IL-4, IFN-γ, IL-17 and IL-10 with ELISA after the 28 days afterwards  Results:7 days, 14days and 28 days after the administration, nasal symptoms of Bimingan group and Loratadine group have been more obviously relieved than the model group(P<0.01). After the model was established,the contents of IL-4 and IL-17 of model group obviously increased and the IFN-γ and IL-10 decreased(P<0.01). Compared with model group, the contents of IL-4 and IL-17 of Bimingan group and Loratadine group decreased and IFN-γ and IL-10 increased.(P<0.01)  Conclusion:It showed that Bimingan Prescription was effective on treating allergic rhinitis. One of the possible mechanism of Bimingan Presciption treating allergic rhinitis was regulating the cell function balance between Th1/Th2 and Th17/Treg through influencing the contents of IL-4, IFN-γ, IL-17 and IL-10.
