薏苡附子败酱散对小鼠急性期溃疡性结肠炎肠黏膜的修复作用

Effects of Yiyi Fuzi Baijiang Powder on intestinal mucosal repair in mice with acute ulcerative colitis

中文摘要:   目的:探讨薏苡附子败酱散对小鼠溃疡性结肠炎(UC)肠黏膜的修复作用。  方法:60只雄性C57BL/6J小鼠随机分为正常组、模型组、美沙拉秦组及薏苡附子败酱散(中药)低、中、高剂量组。除正常组以外,其余各组小鼠均制备成急性期UC模型。于造模第3天起,中药低、中、高剂量组分别给予5.24 g·kg-1·d、10.48 g·kg-1·d、 20.96 g·kg-1·d的薏苡附子败酱散药液,美沙拉秦组给予0.82 g·kg-1·d的美沙拉秦药液,正常组、模型组灌胃给予同体积双蒸水,均持续7 d。实验过程中每日观察小鼠的一般情况,绘制每组小鼠的疾病活动指数(DAI)评分曲线。干预结束后处死小鼠,取结肠组织进行HE染色、组织病理(HI)评分, Western blot检测结肠组织中闭合蛋白(claudin)-1和claudin-2的表达。  结果:模型组DAI评分显著高于正常组,药物干预后美沙拉秦组及中药高、中、低剂量组小鼠与模型组比较,DAI评分明显降低(P<0.05)。HE染色结果显示,正常组小鼠肠组织结构完整;模型组小鼠肠组织结构破坏,黏膜缺损,腺体分离,大量炎性细胞浸润;中药低剂量组病理结果类似模型组;美沙拉秦组及中药中、高剂量组肠组织结构比较完整,部分炎性细胞浸润,HI评分与模型组比较明显降低(P<0.01)。Western blot结果显示,美沙拉秦组、中药高剂量组与模型组比较,claudin-1蛋白表达明显升高(P<0.01),claudin-2蛋白表达降低(P<0.01),且高剂量组较低剂量组作用显著(P<0.01)。  结论:薏苡附子败酱散对急性UC可以改善结肠黏膜的通透性,具有一定的黏膜修复作用。
Abstract:  Objective:To discuss the effects of Yiyi Fuzi Baijiang Powder on intestinal mucosal repair in mice with ulcerative colitis.   Methods:Sixty male C57BL/6J mice were randomly divided into the normal group,model group,mesalazine group and Yiyi Fuzi Baijiang Powder(Chinese medicine)groups with low-,medium- and high-dose. Except the normal group,the acute ulcerative colitis model was established in the other groups. From the 3rd day of modeling,the Chinese medicine groups with low-,medium- and high-dose were treated with decoction of Yiyi Fuzi Baijiang Powder at daily dose of 5.24 g/kg,10.48 g/kg and 20.96 g/kg respectively,the mesalazine group was treated with mesalazine solution at dose of 0.82 g/kg,and the normal group and model group were treated with double distilled water at equivalent volume,with a course of continuous 7 days. During the experiment,the general situation of mice was observed every day,and the curve of disease activity index(DAI) score in each group was drawn. After intervention,the mice were sacrificed,and the colon tissue was collected. The histopathological index(HI) score was evaluated after HE staining,and the protein expressions of claudin-1 and claudin-2 in colon tissue were detected by Western blot.   Results:The DAI score of the model group was significantly higher than that of the normal group.After drug treatment,compared with the model group,the DAI scores of the mesalazine group and Chinese medicine groups with different doses were obviously decreased(P<0.05). The results of HE staining showed that,the intestinal structure of mice in the normal group was intact; while in the model group,the intestinal structure of mice was destroyed,the mucous membrane was impaired,the glands were separated,and there was a large number of inflammatory cells infiltration. The pathological results in the Chinese medicine group with low-dose were similar to those in the model group; while in the mesalazine group and Chinese medicine groups with medium- and high-dose,the intestinal structure of mice was relatively intact,there was a small amount of inflammatory cells infiltration,and the HI score was obviously decreased compared with the model group(P<0.01). The results of Western blot showed that,compared with the model group,the claudin-1 protein expression was obviously increased(P<0.01) and the claudin-2 protein expression was decreased(P<0.01)in the mesalazine group and Chinese medicine group with high-dose,and the effects in the high-dose group were more significant than those in the low-dose group(P<0.01).   Conclusion:Yiyi Fuzi Baijiang Powder can improve the permeability of colonic mucosa in the treatment of acute ulcerative colitis,which shows certain effects of mucosal repair.
