中文摘要: 目的：研究不同浓度土三七水煎剂灌胃法诱导大鼠复制肝小静脉闭塞病(HVOD)动物模型。 方法：土三七2 250 g水煎制成浓度为含生药1.5 g/ml的药液。80只雄性SD大鼠随机分为土三七高浓度组、中浓度组、低浓度组、正常组4组,每组20只,分别给予高浓度(15 g·kg-1·d-1)、中浓度(7.5 g·kg-1·d-1)、低浓度(3.75 g·kg-1·d-1)的土三七药液灌胃,正常组给予温开水灌胃。灌药1周后各组随机处死一半,剩余大鼠于给药2周后处死,采集血清和肝组织,检测血清学指标、观察肝组织病理学变化。 结果：土三七高浓度组、中浓度组和低浓度组大鼠分别有78.95％(15/19)、68.42％(13/19)、57.89％(11/19)出现了HVOD病理表现；土三七高浓度组和中浓度组大鼠谷丙转氨酶(ALT)、总胆红素(TBIL)、总胆汁酸(TBA)均有不同程度显著升高(P<0.01);高浓度组较中浓度组作用更明显(P<0.01)。 结论：土三七水煎剂灌胃法可以成功复制HVOD大鼠模型,其表现和病理改变与人类HVOD相似。
Abstract: Objective:To study the rat model of hepatic veno-occlusive disease(HVOD)induced by gynura segetum(GS)decoction with different concentrations by intragastric administration. Methods: 2 250 g GS was made into the decoction at the concentration of 1.5 g crude drug/ml. 80 male SD rats were randomly divided into the normal group and GS groups with high-,medium- and low-concentration,20 rats in each group.The normal group was treated with warm boiled water by intragastric administration,and the GS groups were treated with the decoction with high-(15 g·kg-1·d-1),medium-(7.5 g·kg-1·d-1) and low-(3.75 g·kg-1·d-1)concentration by intragastric administration respectively. After administration for one week,10 rats in each group were sacrificed,and the other rats were sacrificed after administration for two weeks. The serum and liver tissue were collected. The serum biochemical indicators were detected,and the pathological changes of liver tissue were observed. Results: The incidence rate of HVOD pathological symptoms was 78.95% (15/19) in GS group with high-concentration,68.42％(13/19) in medium-concentration group and 57.89％(11/19) in low-concentration group. The levels of alanine aminotransferase (ALT), total bilirubin (TBIL) and total bile acid (TBA) were significantly increased in GS groups with high- and medium-concentration in varying degrees(P<0.01). Compared with GS group with medium-concentration,the levels of ALT,TBIL and TBA were significantly increased in GS group with high-concentration(P<0.01). Conclusion: The HVOD rat model can be successfully replicated by the gynura segetum decoction with intragastric administration,and its manifestations and pathological changes are similar to those in human with HVOD.
