黄精多糖提取纯化工艺研究

中文摘要:   目的:确定黄精多糖的提取纯化工艺。  方法:以黄精多糖含量为指标,应用蒽酮-浓硫酸比色法测定多糖含量;采用L9(34)三因素三水平正交试验法(加水量8倍、10倍、12倍,煎煮时间30、60、90 min,煎煮次数2、3、4次),确定最佳提取工艺。并在一定条件下,考察AB-8(弱极性)、X-5(非极性) 以及NKA-9(极性) 3种大孔吸附树脂对黄精多糖的纯化效果;研究黄精多糖的纯化工艺。  结果:黄精的最佳提取工艺为加8倍量水,提取3次,每次60 min;NKA-9大孔吸附树脂对黄精多糖的纯化效果最佳,纯化黄精的最佳工艺为20.00 g NKA-9树脂湿法装柱,上样多糖溶液浓度约3.90 mg/ml,上样流速2 ml/min,静置12 h后洗脱,以30%的乙醇溶液120 ml作为洗脱剂,洗脱速度2 ml/min。纯化后的黄精多糖纯度可提高至少20%。  结论:该工艺可行性高,便于操作,适宜批量生产。该研究为国内首次用NKA-9大孔树脂纯化黄精多糖,效果显著,为纯化黄精多糖提供参考依据。
Abstract:  Objective:To determine the extraction and purification technology of rhizoma polygonati polysaccharide.  Methods: With rhizoma polygonati polysaccharide content as index to determine polysaccharide content with the method of colorimetry of sulfuric acid anthrone; three factors and three levels by L9 (34) orthogonal test was used to determine the optimum extracted process; compare AB-8 (weak polarity), X-5 (nonpolar) and NKA-9 (polarity) of three kinds of macroporous adsorption resin's purification effect of rhizoma polygonati polysaccharide under certain conditions; to research purification process conditions of rhizoma polygonati polysaccharide.  Results: The optimum extraction technology of rhizoma polygonati was adding 8 times amount of water, extracting 3 times, both with 60 min each time; NKA-9 macroporous adsorption resin was the best way to purify rhizoma polygonati polysaccharide. The best technology of purification was as follows:wet packing 20.00g resin of NKA-9 into column, about 3.90 mg/ml of sample on polysaccharide concentration with 2 ml/min flow velocity, elute after 12 hours, with 120 ml of 30% ethanol solution as eluent and 2 ml/min elution rate. The purity of rhizoma polygonati polysaccharide could improve at least 20%.  Conclusion:The feasibility of the process was high, with easy operating, suitable for batch production. This is the first time to use NKA-9 macroporous resin to purify rhizoma polygonati polysaccharide in the domestic, it can provide reference for the purification of rhizoma polygonati polysaccharide because of its good effect.
